Yes, laminate flooring works much better if the surface is completely flat. More work, but well worth the effort.

As long as they are smooth and not peeling, cracked or uneven, the laminate will be fine over the tiles.
I hope you are not putting laminate in a kitchen or bathroom. It is a poor choice for those areas because it is easily damaged by moisture. If you want a wood look in those areas, use the TrafficMaster Allure solid vinyl wood-look planks from Home Depot. They are completely impervious to water damage, cost about the same as laminate and, in my opinion, look much better for about the same cost.
